Bio-imaging is a key technique in tracking and monitoring important biological processes and fundamental biomolecular interactions, however the interference of background autofluorescence with targeted fluorophores is problematic for many bio-imaging applications. This study reports on two novel methods for reducing interference with cellular autofluorescence for bio-imaging. The first method uses fluorescent nanodiamonds (FNDs), containing nitrogen vacancy centers. FNDs emit at near-infrared wavelengths typically higher than most cellular autofluorescence; and when appropriately functionalized, can be used for background-free imaging of targeted biomolecules. The second method uses europium-chelating tags with long fluorescence lifetimes. These europium-chelating tags enhance background-free imaging due to the short fluorescent lifetimes of cellular autofluorescence. In this study, we used both methods to target E-selectin, a transmembrane glycoprotein that is activated by inflammation, to demonstrate background-free fluorescent staining in fixed endothelial cells. Our findings indicate that both FND and Europium based staining can improve fluorescent bio-imaging capabilities by reducing competition with cellular autofluorescence. 30 nm nanodiamonds coated with the E-selectin antibody was found to enable the most sensitive detective of E-selectin in inflamed cells, with a 40-fold increase in intensity detected.

The Fudan-UH-MQ Workshop on ‘Nanotechnology meets Bioengineering’ was well supported by CNBP researchers at Macquarie University, Wed 28th June – Thu 29th June.
A joint workshop, organised within the framework of University wide trilateral collaboration between Fudan, Hamburg and Macquarie, the event aimed to enhance collaborations between all three universities as well as generate potential cotutelle PhD candidates.

CNBP was well represented at the 11th International Conference on New Diamond and Nano Carbons, held in Cairns, Australia, 28th May – June 1, 2017.
CNBP Chief Investigator A/Prof Brant Gibson was Co-chair of the conference (pictured) with CNBP researcher Dr Philipp Reineck a contributing speaker, presenting on ‘Bright and photostable nitrogen‐vacancy fluorescence from unprocessed detonation nanodiamonds’.
Also providing a contributing talk was CNBP’s Dr Lindsay Parker, ‘Applications of fluorescent nanodiamonds in cellular molecular tracing.’
Additionally, CNBP’s Andrew Greentree, Ivan Maksymov, Daniel Drumm, Ashleigh Heffernan, Marco Capelli, Nicole Cordina and Emma Wilson gave poster presentations and Brooke Bacon and Desmond Lau provided administrative and technical support respectively.
The conference spanned research topics from fundamental physical and chemical concepts to applied technologically driven applications with carbon based materials. This including single crystal diamond, nanodiamonds, carbon nanotubes, graphene and other carbon nanostructures.

CNBP Research Fellow, Nicole Cordina, has presented a poster at the International Nanomedicine Conference in Coogee (June 27-29).
Her poster presentation “Engineering of Fluorescent Nanodiamonds towards Probing Cytokines in Live Microglial Cells” summarises her recent work with CNBP researcher Guozhen Liu, where 100 nm nanodiamonds were successfully targeted to interleukin-6 on live BV2 cells.
